Thrombosis and Factor Vii Activity

چکیده

The tests for the coagulation time of the blood which are in general use have contributed little to the understanding of abnormal tendencies to thrombosis, as they are extremely insensitive to all but gross changes in the coagulation mechanism. A variety of techniques have been evolved to meet this defect. One group of these methods depends on the addition of an inhibitor, e.g., heparin, to the routine clotting time method (Waugh and Ruddick, 1944; Silverman, 1948). It has been shown, using a heparin sensitized method (Poller, 1956), that statistically the coagulation time is significantly shortened 48 hours after a thrombo-embolic episode. The drawbacks to the use of such methods as simple prediction tests have been discussed (Poller, 1955). As thromboplastin and heparin are to a large extent mutually antagonistic in action, and shortened heparin plasma clotting times have been found in the first few days after operation when tissue damage and other factors may cause an increase in thromboplastin activity, it may be supposed that increased resistance to heparin may be related to the ability to produce thromboplastin in the plasma of such patients. An investigation of various aspects of thromboplastin production was therefore performed in a group of patients and parallel controls. Prothrombin activity, factor V and VII activity, and the production of thromboplastin in the thromboplastin generation test were studied in both groups, together with heparin plasma clotting times. In view of the recently established effect of chylomicra on the coagulation time (Poole, 1955), serum cholesterol values were determined in both groups as a feature of secondary interest.

PREPARATION OF HIGHLY PURIFIED SOLVENTDETERGENT COA GULATION FACTOR VII AND FACTOR IX CONCENTRATES FROM PROTHROMBIN COMPLEX (PPSB)

In this study, anion-exchange chromatography was used to purify factor VII and factor IX from prothrombin complex (PPSB), which contains coagulation factors II, VII, IX and X. For this purpose, DEAE-Sepharose CL-6B gel , Pharmacia column XK-26 , high flow rate and two stepwise gradients with phosphate citrate buffer were used. The yield of the two lyophylized products, factor VII and factor...

Functional and Molecular Characterization of C91S Mutation in the Second Epidermal Growth Factor-like Domain of Factor VII

Background: Coagulation Factor VII is a vitamin K-dependent serine protease which has a pivotal role in the initiation of the coagulation cascade. The congenital Factor VII deficiency is a recessive hemorrhagic disorder that occurs due to mutations of F7 gene. In the present study C91S (p.C91S) substitution was detected in a patient with FVII deficiency. This mutation has not b...

Preparation of factor VII concentrate using CNBr-activated

Background: Factor VII concentrates are used in patients with congenital or acquired factor VII deficiency or treatment of hemophilia patients with inhibitors. In this research, immunoaffinity chromatography was used to purify factor VII from prothrombin complex (Prothrombin-Proconvertin-Stuart Factor-Antihemophilic Factor B or PPSB) which contains coagulation factors II, VII, IX and X. The a...

Factor VII coagulant activity and antigen levels in healthy men are determined by interaction between factor VII genotype and plasma triglyceride concentration.

Ischemic heart disease is caused by a combination of and interaction between a number of genetic and environmental factors. In a study of a group of healthy men from the United Kingdom, such an interaction was identified between the levels of plasma triglycerides and genetic variation determining plasma levels of factor VII, a clotting factor that is associated with risk of ischemic heart disea...

Correlation of vitamin K-dependent clotting factors with cholesterol and triglycerides in healthy young adults.

The plasma level of factor VII activity was a risk factor for the development of ischemic heart disease (IHD) in a prospective epidemiological study of hemostatic factors. We have previously reported significant correlations between factor VII clotting activity or antigen and lipid fractions in a group of 132 young men (< 30 years old) at low risk for IHD and concluded that control of the plasm...

Use of different tissue thromboplastins in the control of anticoagulant-therapy.

As anticoagulant therapy has become more widespread and a greater knowledge has been accumulated regarding the mode of action of coumarin derivatives, problems arising from the use of different thromboplastin preparations have been subjected to greater scrutiny. This paper presents an evaluation of the dependability and significance of different types of tissue thromboplastin in the determinati...
